Located in Burlington, MA, this team is dedicated to developing innovative gene therapies for muscle-related disorders. We are seeking motivated scientists for intern positions at cutting edge biopharmaceutical company. Inclusive environment promoting innovation and teamwork to create gene therapies for rare disease and impact patient lives. 30,000 sq ft. laboratory in beautiful Burlington, MA in a central biotech hub outfitted with state-of-the-art biophysical instrumentation for early-stage R&D and gene therapy process development.
Main Duties & Responsibilities
- The primary responsibility for this role will be supporting research-scale AAV production and characterization. Interns will be trained on all techniques (as needed) and emerge from the role understanding how to make AAV, how to check the quality of the AAV one is making, and how to test the activity of the AAV on cells of interest. Examples of these responsibilities include:
- Genetics: Cloning of genes and engineering of novel viral capsids for tissue/cell type specific trophism. Plasmid production for upstream AAV transfections
- Cell Culture: Adherent/Suspension cells to produce AAVs and evaluate viral infectivity. Bioreactor/shaker flask production experience to produce high titer/high yield AAVs for both cell-based and animal studies.
- Chromatography: Purifications of AAVs using TFF, AKTA pure, and AKTAgo! FPLCs using affinity and IEX resins
- Analytical testing of AAV quality: Digital drop PCR for viral titers/ELISA for capsid titers and percent full vectors, VP ratio characterization, AAV stability assays, potency assays
- Tissue Processing to test AAV function on cells: Tissue homogenization, RNA purification, differential gene expression assays, ELISA, western blot
Qualifications
- Early trainee with relevant degree / degree path in biotechnology or related fields
- Ability to Intern for 6 months or longer
- Familiarity with or experience in molecular and cellular lab techniques
